Discover the Full Details Through Our Olink® Factsheet:

Evotec's partnership with Olink® demonstrates our commitment to accelerating biomarker translation from discovery to clinical application. This synergy reinforces our commitment to advancing precision medicine through robust protein signature analysis.

Olink® technology facilitates this through precise biomarker detection across a wide range of sample types, enabling high-throughput, multiplex protein detection with minimal sample volumes (1 to 2 µL)Olink®'s technology simultaneously measures 21 to 5,000 proteins from minimal sample volumes with exceptional sensitivity.

This technology offers:

  • High-throughput multiplex immunoassays: By analyzing 21 to 5,000 proteins per panel, Olink® can identify disease signatures more quickly and address complex disease mechanisms that require multi-marker profiling
  • High specificity, high sensitivity, and precision: Enables reliable detection of low‑abundance proteins, supporting pharmacodynamic marker discovery and informed patient stratification
  • Only 1 to 2 µL of sample is required: Preserves clinical samples and enables multiple analyses from a single collection
  • Dynamic range from femtograms to nanograms: Quantifies both highly abundant and rare proteins in the same assay without dilution steps, capturing the full continuum of disease‑relevant biomarkers across matrices, from tissue lysates to serum and plasma
  • Scalable technology: Olink® protocols are validated on different matrices, including serum and cerebrospinal fluid, and can be scaled for use in over 20 sample types, preserving consistency from target validation through clinical proof-of-concept studies

Olink® high-plex protein analysis platforms, Explore and Reveal, provide relative quantification of a broad range of proteins at a large scale (Table 1).

Olink® Explore delivers high-throughput profiling of over 5,000 proteins with relative quantification (NPX values). This platform is ideal for unbiased biomarker discovery when you need to cast a wide net across the proteome to identify novel candidates without preconceived hypotheses.

Olink® Reveal profiles over 1,000 proteins, combining broad proteome coverage with deep inflammation profiling. This balanced approach provides more focused analysis than Explore, while offering extensive, in-depth coverage of inflammatory pathways.

Olink® proteomics multiplex biomarker panels are available in TargetFlex, or Focus options, each with different capabilities matched to your biomarker needs (Table 2). The technology uses proximity extension assay (PEA) chemistry to detect proteins in relative and absolute quantification

The Olink® suite of analysis and panel options covers a broad range of proteins relevant to disease areas like oncology, neurology, and inflammation. This means tailored solutions for protein analysis in exploratory biomarker discovery, clinical trial support, and precision assay development.

Case Study: Autoimmune Disease Biomarker Profiling

Autoimmune diseases (AIDs) like IBD, including Crohn’s disease, Lupus, and Multiple Sclerosis share overlapping inflammatory pathways, making it difficult to identify disease-specific protein signatures for patient stratification and treatment selection.

Evotec initiated a dedicated non-interventional clinical trial to access high-quality plasma samples from patients with autoimmune diseases linked to comprehensive clinical data. Evotec performed Olink® Reveal profiling on the samples to identify dysregulated proteins relative to healthy blood samples.

The broad proteome profiling revealed distinct protein signatures across autoimmune conditions. Volcano plot analysis identified proteins with statistically significant differential expression between healthy donors and IBD patients (Figure 1).

NPX (Normalized Protein eXpression) boxplots clearly distinguished disease states from healthy controls, with correlation to clinical data validating the biomarker signatures' relevance to disease severity and patient outcomes.

Olink Reveal proteomics volcano plot and boxplots showing differential protein expression between IBD patients and healthy donors with statistically significant biomarker identification for autoimmune disease patient stratification.

Figure 1. Patient stratification in autoimmune disease using Olink® Reveal profiling. Volcano plot shows differential protein expression between IBD and healthy donors, with boxplots highlighting two significantly dysregulated proteins.

This study demonstrates how Olink®'s high-throughput multiplex approach enables simultaneous profiling of over 1,000 proteins with broad proteome coverage, accelerating the discovery of disease-specific circulating biomarkers that inform precision medicine strategies and clinical trial design for autoimmune disorders.

Evotec collaborates with partners to develop tailored biomarker strategies that span both standalone service projects and fully integrated drug discovery programs.
