Alicia Barker is a Scientist on the Cell Line Development team at Just - Evotec Biologics in Seattle, Washington. She has played a pivotal role in the development of multiple cell lines expressing therapeutic drug candidates for commercial manufacturing. In her current position, Alicia focuses on the design and implementation of high-throughput methods and automation strategies, aimed at expanding the capabilities and efficiency of the cell line development process. Her work contributes to advancing the company’s mission to accelerate and expand access to biotherapeutics.

J.MediaTM: A Concentratable, Room Temperature Stable Perfusion Media For High Biotherapeutic Productivity and Robust Scalability in CHO Cell Culture Platforms
CHO-based Continuous perfusion bioprocessing can offer highly intensified biologics production processes at a fraction of the scale and raw material cost of a traditional fed-batch process but require cell culture media optimization to meet the challenges of high-intensity, long-duration perfusion campaigns. Just - Evotec Biologics has developed a proprietary chemically-defined (CD), animal origin free (AOF), Chinese Hamster Ovary (CHO) perfusion cell culture medium, “J.Media”. Developed to maximize cell viability and titer across scales, it can be concentrated up to 5x and stored at room temperature, which can reduce storage footprint and cold storage requirements for large scale manufacturing campaigns. In addition, the medium can be consumed as either powder or liquid.
J.Media performance was evaluated against leading market perfusion media. J.Media performance was compared against 3 commercially available perfusion media in two J.CHO stable cell lines, each producing separate IgG1 sequences (mAb1 and mAb2). In an 8-day, unbled 24-deep well plate mock perfusion experimental set-up, cells cultured in J.Media exhibited higher volumetric productivity and cumulative titer than the commercially available media tested. We hypothesize that J.Media medium formulations supported better protein production through efficient shuttling of nutrients to protein production cellular machinery as demonstrated by lower specific ammonia and lactate production compared to other evaluated media.
J.Media scalability was further assessed by comparing mAb1 and mAb2 production at 3 L and 500 L scales. Both 3 L and 500 L scale production runs demonstrated similar cumulative titers normalized to bioreactor volume and similar product quality, specifically size variants and glycosylation profiles despite some differences in cell density, volumetric productivity, and viability across scales. These differences are likely due to differences in cell bleed control methods across scales.
Taken together, these data demonstrate Just - Evotec Biologics J.Media provides a scalable, room temperature-stable perfusion medium that should be considered when selecting for high intensity CHO-based perfusion biologics production.
