Expanding Operational Understanding of Single-Pass Tangential Flow Filtration for In-Line Concentration and Diafiltration of Monoclonal Antibodies for End-to-End Continuous Manufacturing
- Single-pass tangential flow filtration (SPTFF) enables a streamlined approach for end-to-end (E2E) continuous manufacturing to perform in-line concentration (ILC) and in-line diafiltration (ILDF) while reducing the costs, labor, and waste of bioprocessing associated with traditional UF/DF.
- E2E continuous processing operates at low mass flowrates and much longer durations relative to comparable batch processes, and the connected nature of E2E requires well controlled flowrates into and between unit operations.
- Generally, the performance capabilities of commercial SPTFF devices are well defined in processes defined as “intensifying batch UF/DF” but are not as well characterized at the low flow regimes characteristic of E2E processing.
- In this study, we screened multiple SPTFF devices to evaluate their performance across both in-line concentration and in-line diafiltration applications for monoclonal antibodies (mAbs). Our findings define an appropriate operational envelope of SPTFF devices and demonstrate their utility and flexibility for mAb processing in E2E continuous bioprocessing platforms.
