Poster summary:
Tregs play a fundamental role in immune homeostasis, and dysfunction or reduction of Tregs is associated with several inflammatory and autoimmune diseases. Currently, a major limitation to generate efficient Treg-targeted treatment approaches is the lack of therapeutic targets to increase Treg immunosuppressive function or numbers. We have developed a modality agnostic CRISPR/Cas9-based screening platform in primary human CD4+ T cells to identify such novel Treg regulatory targets. Validated targets should serve as starting points for drug discovery campaigns in different modalities with clinical relevance for inflammatory and autoimmune diseases.

We believe that allogeneic cell therapies will have a major impact in the field of inflammation and immunology and could provide a curative, single cycle treatment paradigm for several autoimmune diseases. CD19+ B cells and plasmablasts are thought to play an essential role in the onset and progression of systemic lupus erythematosus (SLE). Evotec is developing industrialised GMP manufacturing processes for gene-edited iPSC-derived CD19 CAR NK cells for cell therapy. These could serve as a true off-the-shelf and on-demand treatment option for patients suffering from autoimmune diseases such as SLE.
