Enabling Single Pass Tangential Flow Filtration at Development Scale for Safe and Multi-Condition Experiments With Centralized Data

- Batch tangential flow filtration remains a major bottleneck for highly productive end-to-end continuous processes
- Single pass tangential flow filtration (SPTFF) is an essential tool that allows continuous concentration and diafiltration simultaneously over several days
- Vendors often lag in creating development-scale solutions for these emerging process technologies, which requires scientists to use stand alone equipment for development
- A system to enable SPTFF development has been created by integrating a programmable logic controller (PLC) and auxiliary hardware such as peristaltic pumps, pressure sensors, conductivity sensors, temperature sensors, and an in-line variable path length UV reader at 280 nm
- Custom built human machine interface (HMI) using Supervisory Control and Data Acquisition (SCADA) platform centralizes data from the operation and allows operators to control multiple pumps and monitor pressures, conductivity, temperatures, and protein concentration
- To provide a safety interlock over multiple days of operation, users can configure high-pressure thresholds to automatically stop all pumps when one pressure exceeds its threshold
