Evotec offers both Intact Mass Spectrometry coupled to RapidFire® MS (RFMS) for biophysics screening of covalent modifiers with target proteins, and Native Mass Spectrometry combined with Size Exclusion Chromatography (SEC-MS) for biophysics screening of non-covalent interactions of small molecules with target proteins.
Intact Protein MS Screening Services using RFMS
Screening for covalent modifiers of selected target proteins is enabled by the application of intact protein mass spectrometry. Covalent libraries of up to a few thousand compounds thanks to RapidFire® system that provides fast and efficient analyses of samples and coupled to a MS device suitable for intact MS measurements, such as Time-of-Flight or Orbitrap technologies. The devices are running with 384-well plates. Typical throughput/day is approximately 3,000 samples/device.
A home-built dedicated workflow has been set-up for data analysis coming from high-throughput Intact MS, allowing to determine number of adducts and % binding for each identified hits.
Native MS Services using SEC-MS
Analysis and characterization of non-covalent interactions of small molecules with target proteins is enabled by the application of native mass spectrometry. Direct coupling of Size Exclusion Chromatography (SEC) to MS device suitable for large molecules analysis (Time-of-Flight) allows to analyze proteins in their native shape. After incubation with small molecules, use of this technology can provide direct evidence of non-covalent binding.
Native MS is offering possibility for orthogonal binding characterization of initial hits coming either from functional or affinity assays.
