Cell Line Generation

Evotec offers customized stable cell line development services tailored to your research needs. We generate recombinant cell lines via viral transduction or stable transfection using conventional or transposon-based DNA vectors, delivering rapid-turnaround polyclonal lines or fully validated monoclonal lines with the desired properties.

Each stable cell line is confirmed for target expression using immunoassays or PCR-based analysis (e.g., FACS, Western blot, IF, RT-PCR), and further validated through cell-based assays to ensure functional expression of the target protein or reporter gene. Advanced tools like multicolor flow cytometry, live cell imaging, and high-resolution fluorescence microscopy support deep characterization of protein localization, cell growth behavior, and other phenotypic traits.

Cell Line Development Capabilities

Evotec’s cell line development platform supports both gene overexpression and knockdown approaches using viral vectors or transposon-based gene delivery systems.

  • Custom vector design with flexible promoter and selection marker options

  • Constitutive or inducible expression of target proteins or reporter genes

  • Polycistronic or independent co-expression of auxiliary factors or protein complex subunits

  • Generation of stable cell pools or single clones with tunable expression levels

  • Target expression analysis and functional validation using diverse cell-based assays

Membrane Preparation

Evotec offers tailored cell membrane preparation using scalable, gentle disruption methods to preserve membrane integrity and protein function. We prepare crude or enriched membranes from transient or stable cell lines, optimized for downstream ligand binding assays and target validation.

Membrane Preparation Capabilities

  • Scalable membrane production from up to 5 billion cells per batch
  • Advanced nitrogen decompression systems (e.g., PARR-bomb®, micro-fluidizer) for efficient, non-destructive cell lysis
  • Differential ultracentrifugation for membrane enrichment
  • QC via Western blot, with optional radioligand binding validation
  • Functional characterization through:
    • Filter binding or Scintillation Proximity Assay (SPA)
    • Saturation (Bmax, Kd), kinetic (Koff), and competitive binding (EC50) studies
    • Agonist, antagonist, and allosteric modulator screening

Assay Ready Frozen Cells

Evotec produces high-quality assay-ready frozen cells (ARFs) to support reproducible, scalable, and efficient cell-based assays. Our ARF platform ensures cells are preserved at peak performance, eliminating variability caused by routine cell culture and enabling flexible, on-demand use across high-throughput screening (HTS) and long-term studies.

ARF Cells Production Capabilities

  • Scalable production of up to 10 billion cells per batch
  • Compatible with adherent and suspension cell lines, primary cells, or transiently transfected cells
  • Automated harvesting, aliquoting, and controlled-rate freezing for batch consistency
  • Ready-to-use, reagent-like cell format reduces hands-on time and culture maintenance
  • Batch-to-batch consistency ensures high data reproducibility across time

Ideal for:

  • High-throughput screening (HTS)
  • Quality-controlled long-term assays
  • Parallel cell profiling projects
